Microbots Gene Delivery System Based on Bifidobacteria in a Tumor Model

In cancer, the use of microbots based on anaerobic bacteria as specific transporters targeting tumor tissues has been explored since most solid tumors exhibit hypoxic regions. The aim this study was to develop and characterize magnetic Bifidobacteria iron oxide fluorescent nanoparticles complexed with chitosan a hypoxia inducible plasmid. addition, efficiency for gene delivery evaluated in an vivo model by florescence luminescence. To elaborate microbots, hypoxia-inducible plasmid called nanocomplex (NCs) size 302 nm ζ potential +16 mV were obtained loaded onto membranes. Microbots diameter between 1–2 µm characterized atomic force microscopy (AFM) scanning electron (SEM). injected intravenously through tail vein tumor-bearing mice, then magnet placed focus them area. Forty-eight hours after injection, biodistribution determined greatest luminescence fluorescence emitted found tissue compared normal organs. We created vector that can be directed deliver genes whose expression is regulated microenvironment tumor.
